email alias in To: field

How does the email alias in To field searched and completed? If a user has a
multiple SMTP addresses and one of them is (e-mail address removed) can you just
type nickname only in the to field and it will complete the full name.

I am having an issue where when I put the user's nickname it is not
completing the user's name even after waiting for a while. So, does the
(e-mail address removed) have to be the primary SMTP address?

I am concerned internally only where we have an AD forest with multiple
domains.
