error Help

Whenever I click on the shortcut I get this message;
C:\WINDOWS\SYSTEM32\AUTOEXEC.NT. The system file is not suitable for
running MS-DOS and Microsoft Windows applications. Choose 'Close' to
terminate the application. (With a Close and Ignore button at bottom of
message).

Please tell me how to fix this.
