I enabled instant search and the query builder was working fine. So were
queries of the form "field:value ...". Then my laptop was on the brink of
dying and I had my IT guys copy my files over to a new one. Now instant
search is enabled on my new laptop, but the query builder fields are all
greyed out and queries of the form above don't restrict to the desired fields.

Also, when I click "Search now" from the systray search icon, it pops up a
modal saying, "The folder 'C:\ - Windows Desktop Search' does not exist."
I'm guessing some important data wasn't copied over when I switched machines.
How can I get instant search to rebuild what it needs?

Which version of Windows?
If XP then you could try reinstalling Instant Search. Also check your
Indexing Options in Control Panel and Search Options in Outlook.
