I have recently upgraded to Office 2008 and since then any links contained within an email message do not work.
I will click on the link and a new mail message opens. Can someone please advise what I need to tweak to stop this happening.

Let's eliminate database corruption first.

Drag some test messages to your desktop.
Under Entourage in the Menu bar, select Switch Identity.
Create a new test Identity. No need to enter account info just cancel out.
Drag the test messages into the new Identity.
If they work in the new Identity, this indicates problems in your database.
You need to rebuild.

If the problems still occur, you need to test in a new User. Go to System
Preferences --> Create a New User in Accounts. Switch to the New User by
logging out/in or use Fast User Switching. Do the same test there. Drag the
test messages to Shared folder for access in the new User.

If the problems go away in the new User, the problem exists in your User's
account.

How to rebuild:

Make a duplicate in the Finder first as additional backup. A rebuild does
make a duplicate, but sometimes in the process the original and backup get
merged resulting in total disaster. It's rare but it happens.
