I have a very large column from one spreadsheet that needs to become the header row for another spreadsheet.

How do i do this, except from re-writing the whole lot.

Basically i want all the cells in column B, to now exist in the same order in Row 1.

Select & copy the cells in the one sheet, then in the other sheet select the
starting cell & use Edit> Paste Special - Transpose.

What is "very large"? Remember that Excel 2008 only has 16384 columns.
Anyway, copy the cells in column B. Then select A1. Go to Edit-Paste Special
and check transpose.
