Select the line, then select either the line or pencil tools. Grab the
vertex you want to move.

Unfortunately, when I select the line I want, every other segment in the line
is also selected and any changes I make to the original segment affect the
rest of the line. For example, if I delete the segment, every other segment
is deleted, too.

How do I lengthen or straighten one segment of a line in Visio?

You need to look at the shape's shapesheet.
Select the shape, then menu Window -> Show Shapesheet
You will see several Geometry sections, the formulae here are the key to what you want to
do.

Select the line, choose the pencil tool, click one of the nodes at the end of
the segment you want to change, hold the left mouse button down and drage.
If you want to collapse the segment, once a single node has been identified,
hit the delete key.

You can also do as Paul suggested, if you have both the drawing window open
and the shapesheet open, tile them so you can see both. Then as you click a
geometry entry, the corresponding node in the drawing window will highlight
(black square). Hunt for the nodes you want to delete, then delete the row
in the shapesheet.

Either way works.
